Recombinant HumaXpress™ Cytokines from Human Cells
Cytokines are a group of proteins and polypeptides that organisms use as signaling molecules. Most cytokines are glycoproteins less than 30 kDa in size and bind to specific, high-affinity cell surface receptors. Due to their central role in the immune system, cytokines are involved in a variety of immunological, inflammatory and infectious diseases and widely used in research, diagnostics and therapeutics. Currently, these proteins are predominantly produced in non-human cells (e.g. E coli ) and therefore lack authenticity due to the absence of relevant glycosylation. In addition, a number of important cytokines are not commercially available due to inadequate proteolytic processing or other post-translational modification that occur in the non-human cells.
HumanZyme has developed an efficient human cell-based technology, HumaXpress™, for medium and large scale production of human cytokines. Currently, we have successfully produced more than 60 tag-free cytokines, including difficult-to-express proteins of the TGF beta superfamily. Using GM-CSF, IL-4 and VEGF-165 as examples, we demonstrate our high-authenticity cytokines from human cells can be used as highly preferred reagents for inflammation, cancer, stem cell research, and antibody development.
AAAb™ Antibodies for HumaXpress™ Cytokines
Antibodes for cytokines are routinely raised against proteins produced in non-human cells (e.g. E coli, SF9, CHO ), which have different antigen epitopes due to the absence of physiologically relevant glycosylation and other post-translational modifications. Typically, these antibodies only weakly recognize the natural occurence for critical applications in vivo and ex vivo. HumanZyme has developed the monoclonal and polyclonal antibodies for recognizing authentic human cytokines expressed in human cells. AAAb™ has the advantage of higher affinity and sensitivity for a variety of applications, such as Western Blot, ELISA and Blocking/Neutralization assays.
Ultra-low Endotoxin Recombinant Human from Non-human Cells
Endotoxins are frequent contaminants in the cytokines prepared from bacterial and other expression systems. Even low levels of endotoxins can be toxic to cells or organisms and must be removed. The industry standard reported value for commercially supplied cytokines is <1.0 EU/μg. HumanZyme now offers researchers an expanding range of human cytokines exhibiting ultra-low levels of endotoxin contamination. These cytokines have been prepared to yield 10x to 1000x less endotoxin than the levels reported in standard commercial preparations. Ultra-low endotoxin cytokines are cost effective even for large scale studies.
Recombinant Human Protein Kinases
Protein kinases act on and modify the activity of specific proteins via phosphorylation. These are used extensively to transmit signals and control complex processes in cells. Up to 518 different kinases have been identified in human genome. Due to the critical roles in intracellular communication, dysregulation of protein kinases has been implicated in as many as 400 human diseases including cancer, diabetes, heart diseases, neurological disorders and rheumatoid arthritis. Hence, individual protein kinases are important for drug target validation, protein crystal structure analysis, and drug design and screening. Currently, kinases are predominantly produced in non-human cells (e.g. E coli or insect cells) many of which involve protein truncation and/or in vitro activation, due to the limitations of the expression system.
HumanZyme has developed an efficient human cell-based technology, HumaXpress™ for the rapid production of active recombinant human protein kinases which are full length, in vivo activated, and highly authentic. Currently, we have successfully produced more than 100 active human protein kinases in an engineered human cell line. Using p38α and MK2 as examples, we demonstrate that the properties and inhibition profiles of the human protein kinases produced in human cells are clearly differentiated from versions of the same kinase that were produced in non-human cell systems. Given the high cost and lengthy development of drugs, it would be highly justified to use human kinases with high authenticity for drug screen purpose.
